The neurosphere assay for developmental neurotoxicity testing

چکیده
analogous threshold rules applied, followed by expert intervention for data points for which the automated system failed to generate an expert call. The semi-automated expert calls, which were completed in 5 days, compared to 3 weeks for the manual approach, agreed with the human expert calls for 93% of the 6956 chemicals based on a conservative approach for all combined classified calls. As a proof of concept, the complex nuclear receptor binding/ activation data was successfully transformed into a purposeful dataset. This methodology can be extended to other MIEs, including enzyme inhibition. MIE datasets created this way have useful applications, including faster structural alerts development in a transparent in silico system like Derek Nexus, for insertion in a purposeful database, to support adverse outcome pathway frameworks and for model building.

Developmental neurotoxic effects of Malathion on 3D neurosphere system.
Developmental neurotoxicity (DNT) refers to the toxic effects induced by various chemicals on brain during the early childhood period. As human brains are vulnerable during this period, various chemicals would have significant effects on brains during early childhood. Some toxicants have been confirmed to induce developmental toxic effects on CNS; however, most of agents cannot be identified wi...
متن کاملSpecies-Specific Differential AhR Expression Protects Human Neural Progenitor Cells against Developmental Neurotoxicity of PAHs
BACKGROUND Because of their lipophilicity, persistent organic pollutants (POPs) cross the human placenta, possibly affecting central nervous system development. Most POPs are known aryl hydrocarbon receptor (AhR) ligands and activators of AhR signaling. Therefore, AhR activation has been suggested to cause developmental neurotoxicity (DNT). OBJECTIVE We studied the effects of AhR ligands on b...
متن کاملSeparating chemotherapy-related developmental neurotoxicity from cytotoxicity in monolayer and neurosphere cultures of human fetal brain cells.
Chemotherapy-induced neurotoxicity can reduce the quality of life of patients by affecting their intelligence, senses and mobility. Ten percent of safety-related late-stage clinical failures are due to neurological side effects. Animal models are poor in predicting human neurotoxicity due to interspecies differences and most in vitro assays cannot distinguish neurotoxicity from general cytotoxi...
متن کاملZebrafish as a model for developmental toxicity assessment
The zebrafish embryo has emerged as promising alternative model for traditional in vivo developmental toxicological screening due to their advantageous characteristics as their small size and transparency. In this paper, we reviewed the applicability of the zebrafish embryo model in some relevant areas to human toxicology as developmental toxicity, cardiovascular toxicity and neurotoxicity (beh...
متن کاملDevelopmental neurotoxicity - challenges in the 21st century and in vitro opportunities.
In recent years neurodevelopmental problems in children have increased at a rate that suggests lifestyle factors and chemical exposures as likely contributors. When environmental chemicals contribute to neurodevelopmental disorders developmental neurotoxicity (DNT) becomes an enormous concern. But how can it be tackled? Current animal test- based guidelines are prohibitively expensive, at $ 1.4...
